Abstract:Experimental observation of cryopreservation by means of rapid cooling of pig cornea, which were processed in cryoprotectants, was given in the paper. Combined - staining with alizarin red and trypan blue was used to discriminate the viability of the endothelial cells. Satisfactory results were got and the following conclusion was given: to keep a high percentage of viable endothelial cells, the cryo-processing scheme of high concentation/short time was proved to be effective on reducing the toxicity of cryoprotectants, as the crux of the cooling is to increase the cooling rate of the endothelium, the so called double-stage cooling is preferred.
