Abstract:The antifreeze peptide of bovine bone collagen was prepared from fresh bovine bone and its antifreeze properties were studied. The results showed that the optimum decalcification concentration was 0.25 mol/L and the decalcification time was 48 hours. The maximum extraction rate of collagen was 43.99%. The optimum time for pepsin enzymatic hydrolysis to prepare enzymatic complex was 48 h, the temperature was 25 ℃, and the extraction rate of collagen can reach 48.53%. Meanwhile,the optimum adsorption time of collagen antifreeze peptide by improved ice affinity adsorption method was 4 h, the adsorption temperature was -2 ℃, and the adsorption concentration was 2 mg/ml. Under these conditions, the protein yield was 48.1%, and the thermal hysteresis activity was as high as 5.76 ℃. Bovine bone collagen antifreeze peptide is a kind of protein with high thermal hysteresis activity. It has good application prospects in food freezing, storage and transportation.
