Abstract:In view of different electrical parameters on cervical cancer Hela cells, the self-made pulse power supply and trypan blue were used to study the threshold of electric field intensity for reversible and irreversible electroporation of cells. In particular, the influences of the number of pulses, pulse width and electric field intensity on irreversible perforation rate of cells were investigated. The optimal parameter combination was chosen. The experiments results show that the threshold of reversible electroporation (survival) for Hela cells ranges from 500 V/cm to 750 V/cm with a fixed pulse width of 50 μs and a constant number of 20 pulses. Besides, the threshold of irreversible electroporation (death) ranges from 750 V/cm to 1000 V/cm. In the experiment of optimized parameter combination, the irreversible perforation rate of Hela cells increases with the increase of the number of pluses, pulse width and pulse field intensity, and finally reaches saturation. The irreversible perforation rate of Hela cells is 93.42% when the parameter combination of 1500 V/cm, 60 μs and 70 pulses is selected.
